A reproducible methodology for absolute viral quantification and viability determination in mechanical inoculations of wheat streak mosaic virus

Abstract Wheat streak mosaic virus (WSMV) is a common wheat causing economic losses to production in the Great Plains of North America. Reproducible inoculation WSMV by mechanical methods essential evaluate resistance breeding lines and relies on successful infectivity particles. We used reverse transcription-quantitative PCR (RT-qPCR) for absolute quantification viral genome copy numbers both inoculum infected leaves. A time-course study was designed determine viability over time as well number related phenotypic rating scale. In phosphate buffer, stable with average 1.86 × 10 6 ± 4.85 5 . Plants inoculated this buffer using finger rub contained leaves ranging between 2.66 4 4.69 at 21 28 days post-inoculation. Viral were statistically similar immediately those later points.
